The TissuePress is used as a preliminary step to physical or chemical extraction of intracellular components. It is also used to prepare single cell suspensions. [Narendra P Singh, A Rapid Method for the Preparation of Single-Cell Suspensions from Solid Tissues, Cytometry, Vol. 31, p. 229-232 (1998) and Narendra P Singh, et. al., Exp. Eye Res., Vol. 75, p. 555-560 (2002)]. Operating Instructions Partially retract the piston of the TissuePress to make room for the tissue. Unscrew the brown "cap" and place the tissue sample into the bottom cavity of the Tissue press. If the tissue is partially fibrous or covered with membrane, first trim off the tough parts and/or precut the sample into small pieces. Fibrous tissue will not pass through the TissuePress and can block passage of the soft tissue. Generally, the 2.5 mm sieve is used. If you select the 500 micron stainless steel wire sieve, place it over the press cylinder. Reattach the brown "cap" to the cylinder section. Screw down the piston to extrude the tissue. It will exit the press having a paste-like texture. The yield of pressed, macerated tissue will be 20 - >90%, depending on the amount of connective or fibrous tissue present in the sample.
